Résumé This thesis reports the modification of a bacterial exopolysaccharide (EPS), the EPS HYD 657, from deep-sea hydrothermal origin. This 1.8.106 g/mol polysaccharide is composed of neutral sugars (40-50%) along with acidic sugars (30-40%). The repeating unit is thought to be an undecasaccharide with three small side chains. The native EPS was shown to present biological interest with applications in the manufacture of cosmetics for its abilities to enhance the skin immune defence system. In order to extend its area of interest, we have carried out investigations to obtain low-molecularweight fractions from this high-molecular-weight EPS.

The first part describes the depolymerization of the EPS HYD 657 by a physical way, the ultrasonic depolymerization. The factors the most relevant were determined at a laboratory scale before checking the feasibility and the transposability of these first results at a larger scale (10 L).

The second part deals with a chemical way of depolymerization of our EPS : the free-radical depolymerization with metallic catalysts. A large study was realized in order to determine the influential factors for a better control of the efficiency and the reproducibility of the experiments.

The potential of depolymerized fractions was explored by a screening of biological activities in the field of cosmetic. The results are described in a third part.

The last part of this thesis is devoted to preliminary studies about feasibility of chemical modification of the exopolysaccharide, native or depolymerized. Thus, the addition of charged groups, esterification reactions and oxidation reactions were realized. Some results about characterisation and quantification of the modifications by adapted analysis techniques to each experiment are described.

Résumé en anglais Le travail présenté dans ce mémoire concerne la modification d'un exopolysaccharide (EPS), l'HYD 657, biosynthétisé par une bactérie issue des écosystèmes hydrothermaux profonds. Ce polysaccharide d'un poids moléculaire de 1,8.106 Da est composé de 40-50% d'oses neutres et de 30- 40% d'acides uroniques. L'hypothèse de structure la plus probable décrit son unité de répétition comme un undécasaccharide possédant 3 ramifications courtes. Cet EPS, sous sa forme native, a déjà fait l'objet d'une étude approfondie en vue d'applications dans le domaine de la cosmétique pour ses propriétés apaisantes en application topique cutanée.

Pour élargir le spectre d'applications de cet EPS, la réduction de sa masse molaire a été envisagée.

La première partie décrit la dépolymérisation de l'EPS HYD 657 par une voie physique, les ultrasons. Les facteurs les plus pertinents ont été déterminés à l'échelle du laboratoire avant une première étude d'extension d'échelle afin de vérifier la faisabilité du process à une échelle petit pilote (10 L).

La seconde partie présente une voie chimique de dépolymérisation de notre EPS : la catalyse métallique. Une étude poussée a été réalisée afin de déterminer les facteurs influents pour un meilleur contrôle de l'efficacité et de la reproductibilité des manipulations.

Les résultats des criblages d'activités biologiques des échantillons dépolymérisés dans le domaine visé des applications cosmétiques sont présentés dans une troisième partie.

La dernière partie du mémoire est consacrée à la faisabilité de modifications chimiques de l'EPS, natif ou dépolymérisé. Ainsi, l'ajout de groupements chargés, des réactions d'estérifications et d'oxydation ont été réalisés. Des résultats de caractérisation et de quantification des modifications par des techniques d'analyses adaptées à chaque manipulation sont présentés.
