French abstract Cette thèse avait pour but d‟étudier la biotransformation et les effets génotoxiques de deux HAP d‟intérêt, le benzo[a]pyrène (BaP) et le fluoranthène (Fluo), chez la sole commune Solea solea qui est une espèce benthique sensible à la pollution chimique. L‟utilisation de modèles in vitro, et l‟application de différentes techniques de chimie analytique ont permis de démontrer que, comme chez les vertébrés supérieurs, les dihydrodiols et les phénols sont les métabolites du BaP et du Fluo formés majoritairement par les enzymes hépatiques de soles. In vitro, le potentiel génotoxique du BaP et du Fluo a été démontré chez la sole, par la production d‟adduits et de dommages oxydatifs à l‟ADN. Le Fluo présente une génotoxicité 50 fois plus faible que celle du BaP. Dans le cas du BaP, trois adduits formés chez la sole après activation microsomale ont pu être caractérisés par spectrométrie de masse. Comme chez les vertébrés supérieurs, les adduits formés sont issus de la voie d‟activation des diol-époxydes. Les effets génotoxiques d‟un mélange équimolaire de HAP (BaP, Fluo, Pyr) ont été confirmés in vivo, à l‟échelle de l‟organisme. Une corrélation a été observée entre le niveau de métabolites biliaires de HAP et le niveau de dommages oxydatifs à l‟ADN. Les résultats de cette thèse ont démontré la capacité des enzymes hépatiques de soles à métaboliser le BaP et le Fluo par des voies similaires à celles décrites chez les vertébrés supérieurs. En effet, leur métabolisation se fait par la production de métabolites électrophiles capables de se lier à l‟ADN. Certains métabolites peuvent quant à eux générer des espèces réactives de l‟oxygène responsables de dommages oxydatifs à l‟ADN.

Abstract The aim of this Ph‟D was to study the biotransformation and the genotoxicity of two PAH of interest, namely the benzo[a]pyrene (BaP) and the fluoranthene (Fluo), in the common sole Solea solea, which is a benthic species sensitive to chemical pollution. The use of in vitro exposure models and the application of two different chemical analytical techniques allowed demonstrating that, as in higher vertebrates, the dihydrodiols and the phenols of BaP and Fluo are the major metabolites produced by sole liver enzymes. In vitro, the genotoxic potential of BaP and Fluo was demonstrated in sole, by the production of DNA adducts and oxidative DNA damage. The genotoxicity of Fluo appeared to be 50 fold less important than the genotoxicity of BaP. In the case of BaP, three adducts produced in sole following microsomal activation have been identified by mass spectrometry. As in higher vertebrates, these adducts are produced through the diol-epoxide activation pathway. The genotoxicity of an equimolar mixture of PAHs (BaP, Fluo, Pyr) was confirmed in vivo at the individual scale. A correlation was observed between the level of biliary PAH metabolites and the level of oxidative DNA damage. These results showed the ability of sole liver enzymes to metabolise the BaP and the Fluo through similar pathways to those described in higher vertebrates. Indeed, their in vitro and in vivo metabolism occurs through the production of electrophilic metabolites, able to bind to DNA. Several metabolites are able to generate reactive oxygen species causing oxidative DNA damages.
